Chow served as a member of Parliament for Trinity—Spadina from two thousand six to two thousand fourteen, representing the New Democratic Party (NDP). During her tenure, she was known for her advocacy on various social issues. Her late husband, Jack Layton, was also a significant figure in Canadian politics, serving as the leader of the NDP and the Official Opposition until his passing in two thousand eleven.

In two thousand fourteen, Chow ran for mayor but finished third behind John Tory and Doug Ford. Following this campaign, she took on a new role as a distinguished visiting professor at Toronto Metropolitan University. She made another attempt to return to federal politics in the two thousand fifteen election for Spadina—Fort York but was unsuccessful.
