In fifteen sixty-seven, Mocenigo was a candidate for the dogeship but was defeated by Pietro Loredan. However, following Loredan's death, he successfully ascended to the position of doge in fifteen seventy. His dogaressa, Loredana Marcello, was a noted scholar who passed away in fifteen seventy-two, adding a layer of cultural significance to his reign.

Upon his election, Mocenigo faced immediate challenges as the Ottoman Empire was gearing up for war against Venice.
