He is best known for his remarkable tenure as the governor of South Dakota, where he holds the record for the longest time in office, serving a total of sixteen years. His gubernatorial career spanned two non-consecutive terms, first from nineteen seventy-nine to nineteen eighty-seven and then from nineteen ninety-five to two thousand three, amounting to five thousand eight hundred fifty-one days in total.

Janklow's political journey began when he was appointed as the twenty-fifth Attorney General of South Dakota, a position he held from nineteen seventy-five to nineteen seventy-nine. His career, however, was marred by serious allegations in nineteen seventy-four, when he faced accusations of raping a fifteen-year-old Native American girl, Jancita Eagle Deer, at gunpoint in nineteen sixty-seven. Although he was locally disbarred, the case was later reopened by the FBI but ultimately closed at the request of the White House.

In two thousand two, Janklow was elected to the United States House of Representatives, where he served for just over a year. His political career took a tragic turn in two thousand four when he was convicted of manslaughter following a fatal automobile accident.
