She served as a member of the non-partisan Legislative Assembly of Nunavut, representing the riding of Nattilik from two thousand four until her departure in two thousand eight.

In two thousand eight, Aglukkaq made history by becoming the first Conservative to win the seat in the House of Commons, representing the riding of Nunavut. Her victory marked a significant milestone as she was only the second centre-right candidate to ever secure this position. Aglukkaq's political journey is particularly notable as she became the first Inuk woman to serve in the cabinet, breaking barriers and paving the way for future generations.

Her tenure as a Member of Parliament continued until she faced defeat in the two thousand fifteen federal election against Liberal candidate Hunter Tootoo.
