Šekerinska's political journey began with her leadership of the Social Democratic Union of Macedonia (SDSM), where she was elected president on November fifth, two thousand six, following a no-confidence motion against her predecessor, Vlado Bučkovski. She held this position until the party congress in September two thousand eight, after which Zoran Zaev succeeded her.

In addition to her role as party leader, Šekerinska served as Deputy Prime Minister for European Integration and National Coordinator for Foreign Assistance.
