He embarked on his professional journey with Herfølge Boldklub, which later evolved into HB Køge, making his debut in 2009. His talent quickly caught the attention of larger clubs, leading to a transfer to Brøndby IF in January 2012 for a fee of five hundred fifty thousand euros.

During his time at Brøndby, Makienok faced challenges as the club narrowly avoided relegation. In August 2014, he made a significant move to the Italian club Palermo for a reported fee of two million three hundred thousand euros. His tenure at Palermo included loan spells at English clubs Charlton Athletic and Preston North End, showcasing his adaptability in various leagues.

In 2017, Makienok signed with Dutch side FC Utrecht, where he made two appearances before suffering a knee injury that sidelined him for two years. Following his recovery, he played for Dynamo Dresden and FC St. Pauli in Germany, further honing his skills on the pitch.
