She served as the first lady of Venezuela from two thousand thirteen until two thousand twenty-six, during which time she was married to Nicolás Maduro, the country's president from two thousand thirteen until their capture by the United States in two thousand twenty-six.

Flores's political career includes her role as a deputy in the National Assembly of Venezuela, where she was president from two thousand six to two thousand eleven, representing her home state of Cojedes. In two thousand seventeen, she became a member of the Presidential Commission of the Constituent National Assembly, further solidifying her influence in Venezuelan politics.

On January third, two thousand twenty-six, Cilia Flores and Nicolás Maduro were captured during U.S. strikes in Venezuela. They made their initial court appearance in a Manhattan federal court on January fifth, pleading not guilty to multiple drug trafficking charges. This followed their indictment by the United States District Court for the Southern District of New York in two thousand twenty-one, which sought their extradition on various drug-related offenses.
